Following the withdrawal of Pierce Brosnan, O'Loughlin was a candidate for the role of James Bond, until the role was recast with Daniel Craig in 2005, in preparation for Casino Royale.
Graduated from Australia's National Institute of Dramatic Art in Sydney in June 2002, after completing a full-time three year Bachelor of Dramatic Art program.
Enjoys riding motorcycles, climbing rocks, music, and playing his guitar.
Was a fan of the heavy-metal band Iron Maiden as a kid.
Had a one-year holding contract with CBS regarding his role in Moonlight (2007).
Has a sister, Jennifer, who is one year younger than he is.
His parents got divorced when he was 2-years-old. He was born in the city of Canberra but grew up in Sydney. From the age of 10 onward, he commuted between the two cities to spend time with both parents.
His father is a physics teacher and his mother is a nurse (who also plays the piano).
He was roommates with Jonathan Rhys Meyers in New York City during the filming of August Rush (2007). The two portrayed brothers in the movie.



He did a lot of his own stunt work in Moonlight (2007) and had some training in Kung fu and and Tai chi in order to prepare for his role.

In early March 2012, CBS announced that O'Loughlin would miss shooting some episodes of Hawaii Five-0 (2010) while seeking rehab treatment for dependency on pain medication which had been prescribed to him for a shoulder injury. For several episodes he had only cameo appearances, which were explained as McGarrett being on a covert mission to North Korea.
In August 2013, Los Angeles, California based charity, "Australians in Film" selected him to be honored with their "Breakthrough Award" at a special ceremony to be held in October 2013 in Los Angeles.
